It will be recently-appointed manager Gary Holt's first game in charge following the departure of Steven Pressley to Coventry City.
Falkirk sit third in the First Division after four straight wins, although the side built by Pressley are too far adrift of leaders Partick Thistle to have any chance of winning the title and promotion.
It is the first time the Bairns have come up against top-flight opposition in the tournament this season having beaten Second Division sides Stenhousemuir and Forfar Athletic then Division One rivals Hamilton Academical.
Hibs - without a Scottish Cup triumph since 1902 - return to Glasgow's south side for the first time since their 5-1 mauling by city rivals Hearts in last season's final.
A second successive cup final appearance would be some compensation for the way their Scottish Premier League campaign petered out.
Hibs led the table in November but have won only once in the league since beating champions-elect Celtic in late December.
Hopes of a place in the top six were finally extinguished last weekend with a 3-0 defeat by Neil Lennon's side.
Fenlon's team have, though, come through a tough draw in the Cup, beating three fellow SPL sides, starting with revenge over holders Hearts.
However, they are minus Done after the on-loan Barnsley winger was booked as he made his Hibs debut in their fifth-round win over Aberdeen then collected a second caution in a quarter-final victory at Kilmarnock's Rugby Park.
Hibernian goalkeeper Ben Williams : "We've got players in the squad that have won the Scottish Cup with other clubs and players who have come close.
"So we can learn from that. We know how that affects players and fans, so now it is time to try and put that right.
"When I first moved up here, I was surprised when they told me a club the size of Hibs had gone 111 years without winning the Cup.
"But we know that, if we do win it, we will go down in the history of the club - we will become legends. All players would love that, so that is a great motivating factor.
"Now it's important for us to progress in the Cup, pick up some wins in the last five games of the SPL and make sure that the season doesn't tail off."
Falkirk technical director Alex Smith : "We have won four games in a row and are ready for Hibs.
"Gary has seen our last two games and our players have impressed him, so we can go to the national stadium with confidence, although of course we recognise that we are underdogs.
"Hibs have a terrific striker in Leigh Griffiths, but we have Lyle Taylor, who is a striker that I am sure they will be concerned about keeping quiet."
